What it is
IGF-1 LR3 (Long R3 IGF-I) is a modified variant of insulin-like growth factor I: 83 amino acids instead of 70, with an arginine at position 3 and an extended N-terminus of 13 amino acids. These two modifications reduce binding to IGF-binding proteins (IGFBPs) in plasma, allowing a larger portion of the molecule to remain free in models. Because it is a recombinant protein, it is not listed in the PubChem compound index and this variant does not have a CAS number there.
Research
The analogue was described in the early 1990s in research investigating the significance of IGFBP binding versus receptor binding for the biological activity of IGF-I. It has subsequently been used, among other things, in animal studies on organ growth and as a growth factor in serum-free cell culture. The clinical literature on IGF-I (such as international guidelines for children with growth disorders) pertains to unmodified recombinant IGF-I; the LR3 variant is a research reagent and not a medicinal product.

Lyophilized powder. Proteins of this size are prone to foaming: allow the solvent to run down the inside glass wall and do not shake. Unopened at −20 °C, reconstituted at 2–8 °C. How do I dissolve the powder?
With bacteriostatic water: let the liquid run down the glass wall and swirl gently until everything is dissolved, do not shake. Work aseptically; the reconstitution guide describes the complete protocol. Avoiding foam formation is especially important for a protein of 83 amino acids.

What is the difference between IGF-1 LR3 and regular IGF-1?
IGF-1 LR3 has 13 additional amino acids at the N-terminus and an arginine at position 3. As a result, it binds less to IGF-binding proteins in models; the literature on unmodified IGF-I does not automatically apply to this variant.
Why is there no CAS number listed?
IGF-1 LR3 is a recombinant protein and is not listed in the PubChem molecule index. CAS numbers for this variant do circulate online, but we have not been able to confirm them in a primary database.
What is the shelf life?
Dry and frozen at −20 °C, a vial remains usable for months to years. After reconstitution, it is a matter of weeks, not months: refrigerated at 2–8 °C typically 28–30 days.
